Mapping mutations to the SARS-CoV-2 RBD that escape binding by different classes of antibodies

چکیده
Abstract Monoclonal antibodies targeting a variety of epitopes have been isolated from individuals previously infected with SARS-CoV-2, but the relative contributions these different antibody classes to polyclonal response remains unclear. Here we use yeast-display system map all mutations viral spike receptor-binding domain (RBD) that escape binding by representatives three potently neutralizing anti-RBD high-resolution structures. We compare antibody-escape maps similar for convalescent plasmas, including plasmas whom some were isolated. While plasma are affected across multiple RBD epitopes, plasma-escape most resemble those single class target an epitope on includes site E484. Therefore, although human immune can produce diverse in practice infection is skewed towards already undergoing rapid evolution.

Analysis of Sars-CoV-2 RBD Mutations in Khuzestan Province, Iran—A Retrospective Study, 2021
Background and Objective: One of the main reasons for the ongoing pandemic is the substitutions in the Receptor binding domain(RBD), which is involved in binding to the ACE2 receptor. Methods: Oro-nasopharyngeal swabs were collected from 41 confirmed patients between September 2020 and May 2021 to analyze the mutations of SARS-CoV-2 RBD in Khuzestan Province. Bi-Directional sequencing was use...
متن کاملStability of SARS-CoV-2 in Different Environments and the Effect of Disinfectants on its Survival
In December 2019, the prevalence of a pneumonia with unknown cause was reported in China which was later named Coronavirus Disease 2019 (COVID-19) caused by a virus called SARS-CoV-2. One of the most important ways of COVID-19 transmission is contaminated surfaces. In this review study, we investigated the stability of SARS-CoV-2 in air and on surfaces and the methods of preventing its spread R...
متن کاملDesigning an ELISA Method for Measurement of Human IgG and IgM Antibodies against SARS-CoV-2
Background and purpose: Coronavirus disease 2019 (COVID-19) is a respiratory disease caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). In this study, an indirect ELISA method was designed to measure the human IgM and IgG antibodies against SARS-CoV-2. Materials and methods: Protein sequence of nucleocapsid antigen from SARS-CoV-2 was expressed in E. coli BL21 and then was...
متن کاملWhole-inactivated SARS-CoV-2 vaccines: from fear to hope
Undoubtedly, vaccination can be one of the promising approaches to control infectious diseases such as the COVID-19 pandemic. Inactivated viral vaccines have a history of "vaccine-induced enhanced disease", which may occur when neutralizing antibodies bind to viral antigens without blocking or clearing the infection. This can cause additional inflammation through the mechanisms described for ot...
متن کاملReview on the role of host genetic factors in the susceptibility to SARS-CoV-2
Background: Severe acute respiratory system Cov-2 pandemic has affected the world populations for more than one year. Different incidence and severity of this viral disease among various age range and individuals with different background disease may indicate the pivotal role of host genetic factors in their prevalence and mortality rate. In this regards, present study was performed to review t...
